Aging and immunoglobulin isotype patterns in oral tolerance BJMBR

In the present review we address oral tolerance as an important biological phenomenon and discuss how it is affected by aging. Other factors such as frequency of feeding and previous digestion of the antigen also seem to influence the establishment of oral tolerance. We also analyze immunoglobulin isotypes of specific antibodies formed by tolerant and immunized animals of different ages submitted to different conditions of oral antigen administration. Isotypic patterns were studied as a parameter for assessing the pathways of B and T cell interactions leading to antibody production

Enhanced mucosal re-epithelialization induced by short chain fatty acids in experimental colitis BJMBR

The short chain fatty acids (SCFA) are the best nutrients for the colonocytes. Glucose is poorly used as a fuel but may be transformed into SCFA by colonic bacteria. The aim of this study was to investigate the effect of SCFA or glucose on experimental colitis. Colitis was induced in 30 Wistar rats by colonic instillation of 4% acetic acid. Five days later they were randomized to receive twice a day colonic lavage containing saline (controls, N = 10), 10% hypertonic glucose (N = 10) or SCFA (N = 10) until day 8 when they were killed. At autopsy, the colon was removed and weighed and the mucosa was evaluated macro- and microscopically and stripped out for DNA assay. Data are reported as mean ± SD or median [range] as appropriate. Induced oral tolerance to mucosal-exposed antigens in immunized animals is of particular interest for the development of immunotherapeutic approaches to human allergic diseases. This is a unique feature of mucosal surfaces which represent the main contact interface with the external environment. Most contacts with food protein and microbiota antigens occur at the level of the gut mucosa. In animal models where this natural stimulation is absent, such as germ-free and antigen-free mice, the gut-associated lymphoid tissue (GALT) and systemic immunological activities are underdeveloped. We have shown that food proteins play a critical role in the full development of the immune system. Investigou-se o efeito da suplementação de glutamina na dieta sobre o consumo de ração, ganho de peso e conversão alimentar e sobre a estrutura da mucosa intestinal de frangos. Foram utilizados 320 pintos de corte machos distribuídos em um delineamento inteiramente ao acaso com dois tratamentos e quatro repetições, sendo T1 suplementado com 1% de L-glutamina na dieta e T2 controle. Os índices de desempenho foram analisados aos 7, 21 e 49 dias de idade das aves. Inulin as a growth promoter in diets for rabbits R. Bras. Zootec.

Sixty New Zealand rabbits aged 40 days were divided into 4 groups of 15 animals. The control group received a free diet of antibiotic growth promoter (AGP) and inulin. The second group was supplemented with 2.5 g/kg of inulin. The third was administered AGP with 0.1 g/kg of flavomycin. Finally, the fourth group received a 2.5 and 0.1 g/kg inulin/AGP diet. Body weight gain was higher in the control group. Rabbits supplemented with inulin had lower values ​​of triglycerides compared with the control and AGP groups, and their glucose level was significantly lower than those treated with AGP. La modulación del sistema inmune de mucosas con polisacáridos: Bases para una atractiva alternativa en terapia ABCL

El sistema inmune de mucosas del intestino presenta propiedades únicas: está expuesto a una gran variedad y cantidad de antígenos, desarrolla una actividad inmunológica permanente y mantiene un microambiente fisiológicamente desviado hacia respuestas anti-inflamatorias. Es capaz de distinguir y neutralizar agentes nocivos y reconocer antígenos inocuos, generando entonces un estado de no respuesta llamado tolerancia oral. Este fenómeno natural representa una forma fisiológica, segura e inocua de manipular las respuestas inmunes, para el tratamiento de enfermedades autoinmunes, inflamatorias o alérgicas. Probiotics enhance the recovery of gut atrophy in experimental malnutrition Biocell

AIM: The aim of this study was to evaluate the effect of probiotics on the recovery of the bowel atrophy induced by malnutrition in rats. METHODS: Twenty-and-six Wistar rats (200-250g) were fed with either a normoproteic (sham group, n=6) or a free-protein diet (n=20) during 12 days. Twelve malnourished rats were randomized to recover during 15 days with either a hydrolyzed diet (control group, n=6) or the same diet enriched with probiotics (Streptococcus thermophilus and Lactobacillus helveticus; probiotic group, n=6). RESULTS: Probiotic group showed similar gain of body, liver and bowel weight than controls. Wheat bran- but not oat bran-enriched diets increase the mucosal height of the cecum and colon of newly weaned and aged rats BJMBR

The effect of diets enriched with oat or wheat bran (prepared by the addition of 300 g of each fiber to 1000 g of the regular diet), given for 8 weeks, on the mucosal height of the colon and cecum was investigated. Newly weaned (21 days old) and aged (12 months old) male Wistar rats were used in this study. As compared to controls, diets enriched with wheat bran provoked a significant increase in the mucosal height, whereas oat bran did not cause any effect. In newly weaned rats (21 days old), wheat bran increased the mucosal height (µm) in the cecum by 20% (mean ± SEM for 8 rats; 169.1 ± 5.2 and 202.9 ± 8.0 for control and wheat bran, respectively) and in the colon (218.8 ± 7.2 and 264.5 ± 18.8 for control and wheat bran, respectively).
